No! You can't have 2 backlinks from the same site but there is an exception if lets suppose you made profile backlink and the site has the option to make web 2.0 then in that case you can make the backlinks. Other then that google will consider it as a spam.

We can make multiple links from the same domain or even our domain gets multiple links from the same domain! It won't be considered as spam.
